Physical properties
Each liquid component should be clear and transparent, without precipitation or floccules. The aluminum foil bag of the microplate should be vacuum packed without damage or leakage.
Use
Used to detect the concentration of rat cytotoxic T lymphocyte-associated antigen 4 (CTLA4) in samples such as serum, plasma, and cell culture supernatant.
Experimental principle
This kit uses double antibody sandwich enzyme-linked immunosorbent assay (ELISA). In the microwell ELISA plate pre-coated with anti-rat cytotoxic T lymphocyte-associated antigen 4 (CTLA4) antibody (solid phase antibody), rat cytotoxic T lymphocyte-associated antigen 4 (CTLA4) calibrator and the sample to be tested are added, and then biotin-labeled antibody is added. After incubation and sufficient washing, HRP-coupled avidin is added. After incubation and sufficient washing, the unbound components are removed, and a sandwich complex of solid phase antibody-antigen-biotin-labeled antibody-avidin enzyme is formed on the solid surface of the microwell plate. TMB colorimetric solution is added to produce a blue product, which is finally converted into yellow under the action of the stop solution. The absorbance (OD value) is measured at a wavelength of 450nm on the microplate reader. The absorbance (OD value) is positively correlated with the concentration of rat cytotoxic T lymphocyte-associated antigen 4 (CTLA4) in the sample to be tested. The concentration of rat cytotoxic T lymphocyte-associated antigen 4 (CTLA4) in the sample can be calculated by fitting the calibrator curve.
Specificity
This kit recognizes natural rat cytotoxic T lymphocyte-associated antigen 4 (CTLA4) and has no cross-reactivity with structural analogs.
