Experimental principle
This kit uses double antibody sandwich enzyme-linked immunosorbent assay (ELISA). In the microwell ELISA plate pre-coated with anti-swine 70kDa heat shock protein 1A (HSPA1A) antibody (solid phase antibody), add the swine 70kDa heat shock protein 1A (HSPA1A) calibrator and the sample to be tested, then add the biotin-labeled antibody, after incubation and sufficient washing, add HRP-coupled avidin, after incubation and sufficient washing, remove the unbound components, and form a sandwich complex of solid phase antibody-antigen-biotin-labeled antibody-avidin enzyme on the solid phase surface of the microwell plate. Add TMB colorimetric solution to produce a blue product, which eventually turns into yellow under the action of the stop solution. The absorbance (OD value) is measured at a wavelength of 450nm on the ELISA instrument. The absorbance (OD value) is positively correlated with the concentration of swine 70kDa heat shock protein 1A (HSPA1A) in the sample to be tested. By fitting the calibration curve, the concentration of porcine 70kDa heat shock protein 1A (HSPA1A) in the sample can be calculated.
Specificity
This kit recognizes native porcine 70kDa heat shock protein 1A (HSPA1A) and has no cross-reactivity with structural analogs.
